Nick Watney and Sergio Garcia were tied for the halfway lead at the Barclays Tournament in Farmingdale, New York.
The pair shot rounds of 69 and 68 respectively to lead at eight under, one clear of Vijay Singh and Bob Estes.
Tiger Woods played through a sore back to fire a two-under 69 to sit three adrift, while playing partner Rory McIlroy made the cut by one with a 73.
Opening-day leader Padraig Harrington carded 75 to drop back, denting his chances of a Ryder Cup wildcard.
Harrington who is targeting a seventh Ryder Cup appearance, cannot qualify for one of the 10 automatic places in captain Jose Maria Olazabal's team but may be a contender for a wildcard, although Ian Poulter seems set to be one of two picks for next month's match at Medinah.
But Olazabal, at Gleneagles this week for the Johnnie Walker Championship, said it would take "at least" a victory for Harrington on Sunday to put him in the frame for selection.
Garcia secured his Ryder Cup spot with victory in the rain-hit Wyndham Championship on Monday for his first PGA Tour win since the 2008 Players Championship.
"Golf is a funny game," he said. "When you think that you have it under control, it kicks you down. And then all of a sudden, it gives you something to love it again. If I'm not hitting the shots, then it doesn't help."
Meanwhile, Watney says he is acutely aware of the threat poised by Garcia: "I don't know how Sergio was playing, but he wins last week and he's right up there again this week.
"Golf is funny in that way. Just a couple little things here and there, and all of a sudden you're playing great."
England's Gary Christian, the world number 245, kept in touch with the leaders by shooting 71 to leave him five under for the tournament on Bethpage State Park's notorious Black course.
